Quality
The quality demanded of such amino acids for use
in pharmaceutical applications is prescribed by the procedures
contained in the most commonly used pharmacopoeias (DAB, Ph.Eur,
USP etc.). In addition, when used in biotechnological
applications, particularly for the manufacture of pharmaceutical
active ingredients, further requirements have to be fulfilled with
respect to product properties.

Fractogel EMD media enable all those involved in
bio-pharmaceutical processing the use of a well tested production
tool. Compared to conventional resins, Fractogel EMD media improve
the efficiency of the separation part of the production process.
Yields are higher, processes can be conducted at higher flow rates
therefore reducing throughput time. Because the media last so long
and can be used repeatedly, the economics of the process are very
favourable.
What are Fractogel® EMD process media?
The structure of the Fractogel® particles is
considerably different from that of other hydrophilic
chromatographic resins like dextran, agarose or cellulose.
Fractogel® is a synthetic methacrylate based
polymeric resin providing an excellent pressure stability
resulting in high flow rates. The process media consist of beads
with a particle size between 40 and 90 mm. Fractogel® EMD BioSEC
for size exclusion chromatography has a particle size in the range
20-40 mm. The pores which are formed from intertwined polymer
agglomerates, have a size of about 800 Å enabling a free
diffusion of proteins into the beads. The complete surface is
strongly hydrophilic due to the ether linkages in the polymer.
Long, linear polymer chains
("tentacles") carry the functional ligands. All
tentacles are covalently attached to hydroxyl groups of the
backbone structure of Fractogel®. Thus, both the bead and surface
modification are stable to regeneration and sanitisation. The main
advantage of the tentacle chemistry is the large amount of
sterically accessible ligands for the binding of biomolecules
without any steric hindrance. Target biomolecules are, therefore,
much more tightly bound during the separation process. Different
ligands are utilised for various application areas (ion exchange,
affinity, hydrophobic interaction chromatography).

Work involving biological molecules invariably
involves the use of buffer solutions. The task of the buffer is to
create a defined environment for the biomolecule so that it can be
kept in a stable and native form so that its activity is not
influenced. The buffer itself, however, should not exert any
influence on the biological compound. Equally important, the
buffer should not contain any impurities that may have a negative
effect on the activity or stability of the biomolecule. Hence,
only highly purified substances of defined specification should be
used which guarantee reproducible processes on a lab or production
scale.
VWR International supplies buffers in various
grades. The differences between them rarely involve purity itself
but more the additional parameters that are checked prior to
delivery. For example, whilst the content of nucleases may be an
important quality criterion for biotechnological applications, the
fulfilling of the requirements of a particular pharmacopoeia may
well be critical when it comes to manufacturing a pharmaceutical
active ingredient. However, for certain applications, it may be
desirable to include additional quality parameters not included in
our Certificate of Analysis in the testing programme. Normally it
is possible for this to be done so that a customised product
results.
The same applies to individual package sizes. You
may wish to have a pack containing precisely 11.4kg of sodium
chloride for frequent routine use. We could supply this as a
special pack size. Alternatively you may require a special mixture
of buffers, again in most cases we can oblige. We can produce
mixtures, granulates, ready-to-use solutions and concentrates
according to your wishes or using your procedures – all of
course in strict confidentiality.

Standardised silica gels
Standardised silica gels - The basis for
reproducible chromatography from the laboratory to production.
Merck has specialised over many decades in the
manufacture of standardised silica gel packing materials for
preparative chromatography and offers a wide range of customised
sorbents. Such gels provide the user with unique method
reliability. Regardless of which chromatography process is used,
direct and rapid transfer to a production scale is guaranteed and
without risk. This is ensured by the fact that identical raw
materials are used for all stationary phases, thus providing
identical chromatographic selectivity.
In order to be able to achieve this goal, Merck
produces all silica gel 60 packing materials for analysis, pilot
plant processes and production using the same silica gel raw
material which has specified surface area and a defined specific
pore volume. The selectivity of Merck's silica gels for analytical
and preparative applications is identical.
