100 assays. The novel colorimetric HDAC assay kit provide a simple procedure for fast and convenient measurement of HDAC activity. The assay method eliminates radioactivity/extractions and/or chromatography as used in the traditional assays. The new procedures require only two easy steps; both are performed on the same microtiter plate. First the HDAC substrate which comprises an acetylated lysine side chain is incubated with a sample containing HDAC activity (e.g. HeLa nuclear extract). Deacetylation sensitizes the substrate and thus treatment with the Lysine Developer produces a chromophore which can be analyzed using a plate reader. The assay is well suited for high throughput applications. HDAC inhibitors/substrates and related antibodies are also available separately. |
